Getting started checklist

Is your Institute registered with the Bio21 Collaborative Crystallisation Centre?

  • There is a list on the bottom of the page found here.  This page also describes how to get your institute registered, which involves an official delegate of your institute signing an access agreement.  You can see a sample access agreement by clicking on one of the three categories on the page above.

  • If your institute is registered, then you need to create a user account on the system (https://c3.csiro.au/custRegistration.do)

The user account is defined by the email address: if you want lots of people to be able to see the same images in C3, then you should ask your local IT guru to create a distribution list.  Use that distribution list email address to create the user account for C3, and share the password with the members of the distribution list.

  • Once you are registered, log into the booking software (https://c3.csiro.au/login.do) and open up the Nano Dispenser calendar view by clicking on “Resource Bookings” on the left hand side of the screen.  A link to a printable document which describes the booking form is found here.
  • You need to provide a batchload file for each booking – this file is a text file in a fixed format which is used to upload information about your experiment into Crystaltrak.  You can find a printable document which describes the batchload file here.

If you don’t have any information about your protein at all, we recommend that you set up a single sparse matrix screen at one temperature (JCSG+ N3) to check to see that you are at a reasonable concentration.  As a general rule, you should see 20-40% of your droplets have some type of precipitate after 24 hours if the protein is suitably concentrated.  See the “Novice User Guide”

  • Once you have booked time for your sample to be run, you must make sure that the sample arrives at CSIRO before the scheduled start time for your experiment.  The CSIRO reception desk at 343 Royal Parade is staffed between 8:30am and 4:30pm Monday to Friday.
