1.    Polymer banknote samples in petri dish

2.    Recovery of the dried virus from the banknote samples

00:25 A women in a white biohazard suit sitting in front of a laboratory fume cupboard holding a digital pipette.  Laboratory background air flow noises can be heard.
00:30 Woman speaks – make sure I fix them up.
Background noises continue.
00:49 Camera focusses in on the hands and petri dish. A small drop of pink fluid is pipetted onto one of the square samples. The droplet of fluid is repeatedly sucked up into the pipette and back onto the square sample which effectively recovers the dried virus from the surface of the banknote back into the pink fluid.
The camera moves with the hand and the fluid in the pipette is released into one of the tubes in a multi-well dish.
1:16: Woman speaks “and then we suck it up” referring to the pink fluid being sucked back into the pipette tip. She moves the pipette to the multi-well plate and says “and then we plate it into the first well. Pink fluid is deposited into a tube on the multi-well plate.
Hand moves to the disposal jar and the pipette tip is pushed off the end of the pipette holder.
1:27: “And then we repeat that process to make sure we’ve got as much of the virus off the surface, off the note as we can.”
Pink fluid is sucked up into new pipette tip
1: 33 “And we are doing all these time points in replicate. That is why there are three pieces of note in each petri dish.”
Move to depositing the sample of pink fluid plus re-absorbed virus into the multi-well plate.

3.    Recovery of the dried virus from polymer banknote

01:54: close up vision of a square cut from a five dollar polymer banknote with a circle of dried fluid in the middle of it.
Lab background noise can be heard.
01:59: a droplet of pink liquid is squeezed onto the circle of dried fluid containing SARS-CoV-2.  The fluid is squeezed up into the pipette tip and back out onto the banknote sample several times.
02:11 All the fluid is sucked up into the pipette tip.

4.    Recovery of the dried virus from glass

5.    Virus samples transferred to multi-well plates for analysis

Vision moves to a gloved hand holding a pipette with several tips dipping into a small container of pink fluid.  Lab background noise continues.
03:01 All pipette tips are full of pink fluid and the hand moves to deposit the liquid into a multi-well plate, squeezing a small sample into different lines of wells. This process is repeated. And final shot in this segment is the pipette tips being emptied.

6.    Sterilization of polymer banknote samples
03:45 Camera view is focussed on a hand in green glove holding a plastic test tube with blue lid. The test tube is full of a clear liquid and in the liquid is a pair of plastic tweasers and a cut-up five dollar polymer banknote into small squares.
Women speaks: “And just tell me what they are?”
Another woman speaks: “So, that is the cut-up note in ethanol. This is to sterilize it before we perform the test.”

7.    Working in a biohazard suit & PC4 lab

04:00: Camera view is through a window looking into a Physical containment (PC) level 4 laboratory. A woman in a full biohazard suit and connected to an air supply by blue tubing is sitting on a chair in front of a lab bench. The lab bench is housed within a cupboard that vents the air away.
04:12 Camera zooms in to focus on the digital pipette she is holding. She is setting the volume of liquid that the pipette will measure. Behind her is a yellow plastic screw top waste container.
04:18 Camera zooms out so the whole person in the biohazard suit can be seen.  She is working with the pipette and recovering virus from the polymer banknote samples.
